  代谢性疾病如肥胖症,糖尿病,动脉粥样硬化等的发生和发展机制,主要研究脂肪细胞和肝细胞中的脂肪积累,分泌及分解的分子机制以及与胰岛素敏感性的关系。在细胞水平上研究线粒体的功能,脂滴的形成,融合及与内质网的关系。

  本实验室致力于植物细胞应答过氧化氢的分子机制研究。目前的研究重点:1)植物细胞应答过氧化氢的蛋白质及miRNA的双色网络的构建;2)重要功能蛋白质及miRNA在植物细胞应答过氧化氢反应中的作用解析;3)植物磷酸化蛋白质组及碱性蛋白质分析的方法创新。

  脊椎动物胚胎的分子发育生物学。主要利用斑马鱼做模式动物,分离克隆Nodal和FGF等信号通路的新的介导因子和调节因子,结合分子生物学、细胞生物学、遗传学等技术,研究它们在胚胎发育中的作用及其作用机制。利用诱变技术,鉴定影响早期胚胎发育的母源因子。

  研究兴趣、领域:
   信号传导与细胞骨架:纤毛或真核生物的鞭毛是基于细胞微管的一种亚细胞结构。它不仅起着细胞运动的作用,它还参与动物的发育、细胞分裂和维持各种器官的正常生理功能。纤毛的缺陷会导致多种疾病诸如呼吸道疾病、肾囊肿,内脏转位,肥胖症等。我们利用模式生物衣藻为材料,采用分子生物学、生物化学、细胞生物学等手段,研究纤毛的形成、解聚机理,运动功能、信号传导及在细胞分裂中的调控作用。由于纤毛的高度保守性,我们的研究成果适用于动物细胞的纤毛。我们的发现将对纤毛相关疾病的发病机理的认识、诊断和治疗提供理论上的支撑。
  微藻技术的开发和利用:进行微藻的培养,转基因技术和生物活性物质、生物再生能源的研究。

  生物体蛋白质的 “ 转录 / 翻译 ” 与 “ 降解 ” 是同样重要的命题。人们在 “ 转录和翻译 ” 的研究中取得了众所周知的突破。可以预见, 对蛋白质是如何被特一性降解的研究,必将对生命科学的发展产生重要影响。
  该实验室主要采用植物分子遗传、生物化学、功能基因组和蛋白组学等方法, 研究蛋白降解途径和植物激素调控植物生长发育及抗性的机理。

  研究兴趣、领域
  青蒿素的作用机制:疟疾是危害人类的三大传染病之一。青蒿素是治疗疟疾的良药,也是我国医药科学界对世界医学的重要贡献。青蒿素起效快、副作用小,是至今唯一没有产生抗性的抗疟药物,近来被WHO(世界卫生组织)定为一线抗疟药物。虽然使用多年,青蒿素作用机制是困扰学术界多年的一个问题,至今没有根本解决。
  青蒿素结构独特,没有很好模型解释其作用原理,也没有很好的药物作用机制可以借鉴。我们结合酵母菌、疟原虫以及哺乳类细胞的研究,试图探索青蒿素作用机制这一难题。通过早期工作,我们发现青蒿素通过线粒体达到抑制酵母菌的目的,实验室现在工作是进一步阐述其具体作用方式。
  金属离子代谢和神经褪行性疾病:金属离子代谢和疾病的关系:神经褪行性疾病包括老年痴呆或阿尔致海默(AD)、帕金森(PD)等常见中老年人疾病,随着社会的年龄老化神经褪行性疾病越来越成为家庭以及社会的负担。金属离子催化很多氧化还原反应,是细胞内关键催化剂,也是很多自由基产生的催化剂。 神经褪行性疾病伴随金属离子代谢异常,但这种因果关系至今不明。 我们利用果蝇作为模式生物试图回答金属离子在神经褪行性疾病或其它疾病发生、发展中的作用,为开拓新的疾病治疗方向提供依据。
  酵母死亡的基因调控:酵母菌作为模式生物过去很少用于程序性死亡研究,近来发现这一过程在单细胞酵母中也具有类似或保守的现象。过多的金属离子引起细胞死亡,我们试图系统分析这种死亡是由什么基因控制的。我们希望利用酵母菌强有力的遗传手段获得细胞死亡的新知识。

  1968年出生于山东;1993年就读于英国剑桥大学并获博士学位;1997年在美国华盛顿大学(St. Louis)生物系从事博士后研究;2001受聘于瑞士Friedrich Miecher生物医学研究所担任研究组组长(助理教授);2005年入选清华大学“百人计划”,清华大学医学院担任教授、博士生导师; 2006年回国并获国家自然科学基金委的“国家杰出青年科学基金”;2007年被聘为教育部生命科学(表观遗传学)“长江学者”特聘教授。现担任清华大学医学院学术委员会主任,国家自然科学基金委员会表观遗传重大项目专家组成员(兼副组长)等。
  实验室目前主要研究方向:
  1)干细胞分化与个体生长发育的表观遗传学;2)癌症形成的表观遗传调控网络。研究采用的动物模型包括果蝇、小鼠及细胞模式系统(如干细胞和肿瘤细胞)。

  1989年3月于西北农业大学获博士学位。1995年4月至1998年10月在美国圣路易斯大学医学院、华盛顿大学医学院和伯明翰大学医学院做访问学者和博士后。1998年10月回国来清华大学生物系工作。现为清华大学医学院教授、博士生导师,清华大学生命科学院校内兼职教授,FEBS Letters杂志编委、《解剖学报》编委。主要从事细胞信号转导与疾病的关系研究

  重要信号蛋白的泛素化降解调控;STAT3蛋白活性调控;TGF-β,Wnt,FGF等信号在细胞增殖与分化中的作用;肿瘤转移的基因调控;IL-17在炎症反应中的作用。发表SCI收录论文40多篇,获发明专利17项,其中1项美国专利。发表论文获中国细胞生物学学会全国代表大会优秀青年科技论文一等奖1项,三等奖4项。回国后,承担了十数门次课程的教学任务,《热爱教育事业,积极从事教学工作,努力提高教学质量》获2002年清华大学教学工作优秀成果一等奖。

  1978至1982年在北京医科大学基础医学系学习,获医学学士。1985年获北京医科大学免疫学硕士。1987至1991年在澳大利亚墨尔本大学医学生物学系学习,获博士学位。1991至1995年在澳大利亚墨尔本霍尔医学研究所(The Walter & Eliza Hall Institute of Medical Research)从事博士后研究。1996至2000年任澳大利亚墨尔本霍尔医学研究所研究员。2000年获国家自然科学基金海外青年合作基金(杰出青年基金B类)。2001年入选北京大学医学部,教育部长江学者讲座教授。2001至2009年任澳大利亚墨尔本霍尔医学研究所高级研究员,研究室主任。2009年任清华大学教授,并入选国家“千人计划”。主要科研领域包括运用细胞及分子免疫学方法研究免疫细胞发育与功能的调节及其在免疫相关疾病发病中的作用机理。

  李海涛  博士,特别研究员
  教育工作经历
  1993年9月-1997年7月,在山东大学微生物系微生物学专业学习,获学士学位;1997年9月-2003年6月,在中国科学院生物物理研究所学习,获博士学位。在读博士学位期间,师从生物物理研究所中国科学院院士常文瑞教授,主要从事了亚含铜硝酸还原酶及人源磺酰基转移酶的结构与功能研究。在阐明亚硝酸还原酶四级结构与酶活关系以及人磺酰基转移酶的底物识别分子机理等方面做了深入而系统地分析。其间以第一作者身份在国际期刊发表四篇专业论文,2002年参加中国生物物理学会第九届年会获得“最佳墙报奖”,2003年获得中国科学院“地奥”奖学金。
  留学美国期间,在纽约Memorial Sloan-Kettering Cancer Center美国科学院院士Dinshaw Patel实验室从事博士后研究,并于2006年晋级为该中心高级研究员(Senior Research Scientist)。主要从事表观遗传学领域的结构生物学研究,在阐明与白血病,癌症等疾病密切相关的蛋白质分子“组蛋白密码”识别机制,以及RNAi介导的异染色质结构调控等方面取得了颇有影响的原创性成果。目前已在相关领域发表专业论文十二篇,其中包括《自然》,《分子细胞》等,研究成果迄今已被同行引用500多次。2007年应邀为《自然评论-分子细胞生物学》以及《自然-结构分子生物学》撰写表观遗传学领域进展综述,并参与提出以核小体为基本单位的“组蛋白密码”多价态识别假说。同年得到Abcam公司资助,为《自然评论-分子细胞生物学》主导制作了“组蛋白密码与识别”科普海报。
